TopicSelective polymer therapeutics for eradication of cancer stem cells along with deep view of intracellular communication by exosomes and microvesicules
AnnotationPolymeric drug carriers allow the contolled drug targeting to the target site together with minimizing the side effects of therapy. The cancer stem cell remains a population resistant to the treatment of both conventional and polymeric drugs, and often is the origin of metastases. The focus on these types of cells is one of the unresolved topics of tumor therapy.Hence, selective drugs for the treatment of tumor stem cells are tested, and in vitro models are developed for their study using spheroids. To monitor the modulation of the body's response to anticancer therapy and the development of metastases, exosomes and microvesics have been studied in recent years. These vesicles derived from the plasma membrane of the cells are important indicators of physiological and pathological processes. Studying cell response with a focus on tumor stem cells and characterization of tumor tissue by signaling microparticles after targeted treatment with polymeric drugs can significantly improve the development of anticancer drugs. The aim of the project will be the preparation of polymeric drugs, the selection of tumor stem cells and the study of cellular response on interaction with polymeric carriers and drugs, respectively, respectively intercellular communication via microparticles and exosomes, their characterization and modulation of their formation depending on the type of carrier and drug. The proposed theme concerns the overlapping fields of biochemistry, macromolecular chemistry and molecular biology, which is currently highly topical in the light of the necessary interconnection of leading to a more efficient drug development and therapeutic approaches. The applicant will be trained in principal techniques of polymer preparation as well as in principal methods of cell and molecular biology.
